چکیده

Denne afhandling er af Danmarks Farmaceutiske Højskole antaget til offentligt at forsvares for den farmaceutiske doktorgrad. For Anita, without whose support this would have been much harder. Preface i Preface No Man is an Island. This immortal phrase by John Donne also holds true when it comes to science. Thus the present thesis would not have been possible to write without the many fruitful collaborations I have enjoyed during my research. First of all I would like to express my gratitude to Professor Povl Krogsgaard-Larsen who has been of paramount importance to my research due to his many helpful suggestions and his endless support. I would also like to thank all present and former members of the NeuroMedicinal Chemistry group for their contributions, in particular the co-authors of the papers forming the thesis are gratefully acknowledged. Also I extend my thanks to Heidi Petersen and Anne Nordly for excellent technical and secreterial assistance, respectively. Finally, I thank Drs. Spalding for fruitful discussions and collaborations and Dr. Richard Mountfield for helpful suggestions regarding the thesis manuscript. G-protein Coupled Receptor Dimerization

A growing body of evidence suggests that GPCRs exist and function as dimers or higher oligomers. The evidence for GPCR dimerization comes from biochemical, biophysical and functional studies. In addition, researchers have shown the occurrence of heterodimerization between different members of the GPCR family. Two receptors can interact with each other to make a dimer through their extracellular...

High glucose condition down-regulates the inhibitory G-protein subunit, Gαi, in pheochromocytoma PC12 cells

Introduction: G-proteins have an important role in the cell signaling of numerous receptors. The situation of G-proteins in health and disease and their critical role in the development of diabetic side effects is an interested scientific field. Here, the changes in the expression of G-protein subunits (Gαi, Gαs and Gβ) were evaluated in hyperglycemic situation of PC12 cells as...

Metabotropic glutamate receptors and their ligands applications in neurological and psychiatric disorders

Metabotropic glutamate receptors (mGluRs) consist of a large family of G-protein coupled receptors that are critical for regulating normal neuronal function in the central nervous system. The wide distribution and diverse physiological roles of various mGluR subtypes make them highly attractive targets for the treatment of a number of neurological and psychiatric disorders. The discovery of ...

Preparation and biochemical characterisation of nanoconjugates of functionalized carbon nanotubes and cytochrome c

Objective(s): The present work deals with the preparation of nanobioconjugates based on the immobilization of cytochrome c (cyt c) on functionalized multi-wall carbon nanotubes (f-MWCNTs). The effect of the nanosupport and the immobilization procedure on the biochemical and structural characteristics of the immobilized protein was investigated. Methods: </strong...

A Mimic of the Tumor Microenvironment on GPR30 Gene Expression in Breast Cancer

Introduction: The G-protein coupled receptor 30 (GPR30) gene is a member of the G-protein coupled receptor (GPCR) family; involved in breast, endometrial, and ovarian cancers. Many GPCR receptors that are implicated in several types of human cancers are correlated with increased cell proliferation and tumor progression; especially GPR30 gene. Methods: The breast cancer MCF-7 and MDA-MB-231 cel...

Pharmacological Characterisation of the hP2Yn and XlP2Yn receptors

Extracellular nucleotides and nucleosides are important signalling molecules that exert a diverse range of physiological responses throughout the body. These chemical messengers often transduce their effects through interaction with specific cell surface receptors called purinoceptors. The P2Y purinoceptor family binds extracellular nucleotides, principally ATP, ADP, UTP and UDP. Conformational...
